Comparison of RAPD, RFLP, AFLP and SSR markers for diversity studies in tropical maize inbred lines Genet. Mol. Biol.

In order to compare their relative efficiencies as markers and to find the most suitable marker for maize diversity studies we evaluated 18 inbred tropical maize lines using a number of different loci as markers. The loci used were: 774 amplified fragment length polymorphisms (AFLPs); 262 random amplified polymorphic DNAs (RAPDs); 185 restriction fragment length polymorphisms (RFLPs); and 68 simple sequence repeats (SSR). For estimating genetic distance the AFLP and RFLP markers gave the most correlated results, with a correlation coefficient of r = 0.87. Bootstrap analysis were used to evaluate the number of loci for the markers and the coefficients of variation (CV) revealed a skewed distribution. Se determinó la estructura de la diversidad genética y el patrón de cruzamiento en poblaciones naturales de Pseudotsuga menziesii (Mirb.) Franco en México con base en información de isoenzimas. Para evaluar la diversidad genética se utilizó una muestra de 11 poblaciones naturales distribuidas en diferentes regiones geográficas (noroeste, noreste y centro) del país; el patrón de cruzamiento y su efecto sobre el nivel de endogamia sólo se evaluó en una muestra de tres poblaciones, representativas de cada región geográfica. Evaluation of the genetic diversity among maize lines (Zea mays L.) by DNA fingerprinting analysis Ciência Rural

Informativo DNA fingerprint profiles of eight homozygotic maize lines were obtained by the electrophoretic separation of DNA restriction fragments and the ir hybridization with the minisatellite probe R18.1. The analysis of the bandsharing frequencies allowed to identify all the lines and to estimate the genetic distances between them. The relationship obtained by DNA fingerprinting analysis of the eight inbreed lines was highly consistem with the ir genetical origin.

Genetic distance and its association with heterosis in cacao BABT

The efficiency of cacao breeding program can be increased by choosing superior crosses to be made between divergent clones. We assessed the genetic distance among five clones with RAPD data (genetic distance - GD) and with yield component data (Mahalanobis distance - MD). The clones were evaluated in a diallel, during five years, for five yield components. A total of 130 RAPD bands were scored. GD and MD were used to determine the correlation between genetic distances among clones and the performance of their hybrids. The correlation between GD and MD was 0.67 (P=0.03). Genetic diversity is essential for any breeding program. However, breeders tend to concentrate on specific genotypes, which combine traits of interest and may be used as progenitors in several breeding programs. Common bean (Phaseolus vulgaris L.) breeding programs are not different in this sense. In this study, the genetic diversity of 21 common bean elite lines from the Bean Regional Trials conducted by the Embrapa Rice and Bean Research Center was evaluated using the Random Amplified Polymorphic DNA (RAPD) and pedigree analyses. Clustering and ordination procedures in multivariate analyses have been widely used to describe patterns of genetic distances. However, in some cases, such as when dealing with Jaccard coefficients based on RAPD data, these techniques may fail to represent genetic distances because of the high dimensionality of the genetic distances caused by stochastic variation in DNA fragments among the units analyzed (species or populations). Kimberleytrachia Köhler, 2011 is a genus of camaenid land snail endemic to the Western Australian Kimberley region. It comprises twelve previously recognised species, all of which occur within the high precipitation zone along the north-western coast between the Admiralty Gulf and King Sound and within less than about 50 km distance from the coast.
